چکیده

As part of a study on enzymic catalysis of group transfer, this laboratory has previously reported on the enzymic transfer of the glucuronyl radical from a variety of donor substrates to a number of acceptor alcohols (1). The phenomenon was studied in detail and conditions for efficient group transfer were investigated. The present paper reports the use of a similar approach in studying the acid phosphatase-catalyzed transfer of the phosphoryl radical from suitable donor substrates to acceptor molecules. Conditions for efficient transfer and the nature of the phenomenon are described herein. The comparison of phosphoryl and glucuronyl transfer provides points of similarity and of difference which are dealt with in the discussion.

The stereochemical course of phosphoryl transfer catalysed by glucose 6-phosphatase.

Rat liver microsomal glucose 6-phosphatase catalyses phosphoryl transfer between D-glucose 6-[(R)-16O,17O,18O]phosphate and D-glucose with retention of configuration at the phosphorus atom. Since individual phosphoryl-transfer steps appear in general to occur with inversion of configuration, this observation is most simply interpreted in terms of a double-displacement mechanism with a phosphory...

Studies on possible phosphoryl enzyme formation in catalysis by hexokinase, pyruvate kinase, and glucose 6-phosphatase.

One possible mode of enzymic phosphoryli transfer involves formation of phosphoenzymes as intermediates. Thus far, however, convincing evidence for the formation of such intermediates has been obtained only for the phosphoglucomutase reaction (I, 2) and, to a lesser extent, for the phosphoglyceromutase reaction (3). Suggestive evidence has been reported for the participation of phosphoenzyme in...

Catalysis of the Hydrolysis of Phosphorylated Pyridines by Mg(OH)+: A Possible Model for Enzymatic Phosphoryl Transfert

The second-order rate constants for reaction of the Mg2+ complexes of phosphorylated pyridine monoanions with Mg(OH)+ a re 104-106-fold larger than the second-order rate constants for their reaction with water (25 OC, ionic strength 1.5). Of the 106-fold rate enhancement with the phosphorylated 4morpholinopyridine/Mg2 complex, 1 04-fold is attributed to the greater nucleophilicity of Mg(OH)+ co...

Arginine coordination in enzymatic phosphoryl transfer: evaluation of the effect of Arg166 mutations in Escherichia coli alkaline phosphatase.

Arginine residues are commonly found in the active sites of enzymes catalyzing phosphoryl transfer reactions. Numerous site-directed mutagenesis experiments establish the importance of these residues for efficient catalysis, but their role in catalysis is not clear. To examine the role of arginine residues in the phosphoryl transfer reaction, we have measured the consequences of mutations to ar...

Anionic polymers. I. Inhibition of prostatic acid phosphatase.

In the course of synthesizing a number of aryl phosphate esters as substrates for prostatic acid phosphatase, polymeric phosphorylated products obtained by reaction of 4,4’-dihydroxybiphenyl with phosphorus oxychloride were found to be potent enzyme inhibitors. This report describes the preparation of these anionic polymers and some characteristics of their inhibition of prostatic acid phosphat...
